Protective effect of Pu Yuan and stomach endotoxemia in rats with acute gastric mucosal lesion

OBJECTIVE: To investigate the protective effect Pu Yuan and stomach capsule on endotoxemia model rat gastric mucosa. Methods 30 rats were randomly divided into control group and intervention group. 15 rats in each group, the intervention group to 1.0g / kg Pu Yuan and gastric gavage once daily, control group received the same amount of normal saline. 7 days after the two groups were intravenously injected endotoxin establish endotoxemia model. After 48 hours the animals were killed, the MDA content in gastric tissue. Parallel gastric ulcer index assessment. The intervention group gastric MDA content and assessment index was significantly less than the control group (P <0.05). Conclusions in the rat endotoxemia Pu Yuan and the stomach protective effect on the gastric mucosa.

Results The MDA and gastric ulcer indexof intervention groupwere significantly lower than those of control group.Conclusion PYHW treatment could exert the protective effect in endotoxemia ofrats.

Pu Yuan and stomach by Yuan Hu, sweet with a group of 6 herbs from the Chinese medicine preparation, good for the stomach and peptic ulcer treatment. The experiments were performed on the basis of the establishment of the rat model of endotoxemia observation, Pu Yuan and mucosal protective effect, so as to provide some theoretical basis for clinical infection with severe trauma and cancer patients with adjuvant therapy.

MATERIALS AND METHODS

1.1 Materials

1.1.1 Animal 30 SP rats weighing at 250g ~ 300g, were purchased from Experimental Animal Center of Shandong University School of Medicine.

1.1.2 Pu Yuan and stomach capsules purchased from Qingdao Huaren Pharmaceutical Co., Ltd. large.

1.1.3MDA kit purchased from Nanjing Jiancheng Bio Limited.

1.1.4 toxins within 055B5 purchased from sigma company.

1.2 Method:

1.2.1 Animal grouping and model 30 rats were randomly divided into control and intervention groups of 15 rats. Pu Yuan and intervention group were given capsules stomach aqueous solvent 1.0g / kg gavage, the control group was given normal saline orally once daily for 7 days. 7 days after the two groups of animals under anesthesia via the right jugular vein central venous catheter, giving injections of E. coli endotoxin (endotoxin O55B5) 1.0mg / kg intravenous injection, rectal temperature kept about 37 ℃.

1.2.2 Determination of gastric tissue MDA: anatomy experiment rats take gastric tissue, dry water, add 2 ml of PBS solution to cut into pieces, homogenized mechanism into 10% of the tissue, 4 ℃ low temperature centrifugation 5000 r / min, 15 minutes, the supernatant -20 ℃ refrigerator tested. The method of operation according to kit assay MDA with thiobarbituric acid

1.2.3 standard calculation according to Guth gastric ulcer index score, including ① punctate ulcers and erosions, ulcers calculated every three point one point; ② strips bleeding, 1mm elderly calculated 1 minute, 2mm elderly calculation 2 points , 3mm elderly calculate 3 points. Sum of the two exponential rates

1.2.4 gastric tissues were fixed, dehydrated, embedded, sliced observe pathological changes by HE staining under the microscope.

1.3 The statistical procedure

All data were expressed as mean ± standard deviation recorded between the two groups of data using t test, P <0.05 was statistically significant.

2 Results

2.1 gastric tissue MDA: intervention group after Pu Yuan and stomach gastric gavage MDA content was significantly lower than the control group (P <0.05)

2.2 ulcer index score: the intervention group than in the control group was significantly reduced (P <0.05)

2.3 pathology, two groups of animals were killed after gastric mucosa observed, gross pathology were edema, erosion control group, the intervention group compared with heavy bleeding. Endoscopic submucosal inflammatory cells are infiltrated, mucous layer cell edema, loss, destruction of the gland.

 3 Discussion

    Pu Yuan and stomach capsule is a pure Chinese medicine, the main group is the British public square, Yuan Hu, Wulingzhi Chen chips and licorice, with its pharmacological effects as anti-inflammatory sterilization, blood circulation and detoxification. In a related study showed that Pu Yuan and gastric mucosal injury for ethanol, trauma and other factors have a protective effect [1]. In clinical medicine for gastritis, gastric ulcer and has good effect.

    Endotoxemia more due to severe infection, bleeding, shock, trauma, and tumor-induced, are common in critically ill patients [2], high mortality clinic. Endotoxemia can cause acute gastric mucosal lesions, gastrointestinal bleeding, according to reports on the cause of approximately 20% [3], we have a higher mortality rate. The majority of patients with acute erosive gastritis in ICU rescue therapy, or even gastrointestinal bleeding and worse, even death. So for acute erosive gastritis treatment to improve the prognosis of patients has important significance [4]. In this experiment successfully established mouse model of endotoxemia in the stomach and gross pathology seen: gastric hyperemia erosion accompanied by bleeding, stomach and intervention group MDA and gastric ulcer index assessment was significantly lower than the control group. Thus, Pu Yuan and stomach mucosa capsule endotoxemia in a positive protective effect.
